Tumor Area Quantification

Tumor Area Quantification

Molecular pathology is becoming more and more important in today's pathology. One of the main challenges of any molecular test is that it can reliably detect mutations in a sample composed of a mixture of tumor cells and normal cells, especially when the tumor content is very low. The percentage of tumor cells is usually estimated by a pathologist, which is important for the final diagnosis and prognosis of cancer.

Tumor Area Quantification

Microscopic examination of tissue morphology using H&E is a standard practice for visual analysis of tissue specimens in histopathology. The percentage of tumor cells is an important indicator to evaluate the total area of the tumor. CD BioSciences uses H&E to stain pathological tissues to distinguish tumor cells from non-tumor cells, then counts tumor cells and quantitatively evaluates tumor area.

Analysis method Brightfield imaging
Sample requirement Whole tissue fixed and stored in PBS with 0.05% azide
Formalin fixed paraffin embedded (FFPE) tissue blocks
Tissues embedded in OCT
Pre-stained and mounted slides
Digitized slide images
Markers Hematoxylin and eosin
Image analysis Cell counting
Tumor area quantification
Analysis of nuclear morphology

Tumor Area Quantification Workflow

The following is the general workflow of tumor area quantification:

Step 1

Sample preparation

Tissue samples are transferred to PBS w/0.05% azide or made into FFPE, OCT compounds.

Step 2

Sample staining

The samples were processed, sectioned and stained with hematoxylin and eosin.

Step 3

Imaging

The sample slide is imaged under a high-throughput slide scanner.

Step 4

Analyze

Analyze the images according to customer requirements.

Step 5

Send data

Send images and quantitative reports to customers.

Delivery

Whole slide images in RGB format
ROI masks (tumor and non-tumor regions)

Data tables containing cell counts
Other relevant data
